    高效毛细管电泳法测定加杨叶芦丁含量的动态变化

    Determining the dynamics of rutin content in Populus×canadensis Moench. leaves by high performance capillary electrophoresis

    • 摘要: 建立了加杨叶中芦丁含量测定的高效毛细管电泳方法。用甲醇溶解样品采用超声提取,研究了缓冲溶液浓度、pH值、电压、温度及进样时间等因素对分离检测的影响;确定了最佳的测定条件: 50 mmol/L硼酸盐缓冲溶液(pH值为9.3),压力进样时间为 5 s,温度为25℃,运行电压为20 kV,检测波长为254 nm。在此条件下,峰面积与芦丁浓度具有良好的线性关系,平均回收率达到 98.58%,方法准确。同时,利用建立的方法研究了加杨叶生长过程中芦丁含量的变化。

       

      Abstract: The method for determining rutin content of Populus×canadensis Moench. leaves was developed by high performance capillary electrophoresis(HPCE). Sample was extracted with methanol by ultrasonic.The optimum determination conditions were investigated experimentally and the results were as follows: the running buffer of 50 mmol/L borate (pH=9.3), injection time with pressure of 5 s,temperature of 25℃,applied voltage of 20 kV,detection wavelength of 254 nm. Under the optimum conditions, a good linearity between peak area and concentration of rutin was found.The average recovery is 98.58% and the method is accurate. Meanwhile, the developed method was utilized to investigate the variation of rutin content during the growth of P.×canadensis leaves.
