    Effects of NaCl stress on activity and expression of plasma membrane H+-ATPase in Broussonetia papyrifera

    • 摘要: 以构树幼苗根组织和叶组织为实验材料,研究了NaCl胁迫对质膜H+ATPase活性和表达的影响。结果表明:盐胁迫促进了构树幼苗质膜H+ATPase活性的提高。随着NaCl处理浓度的升高,根中质膜H+ -ATPase的活性呈持续上升趋势,而叶片中表现为先升高后降低的趋势。激光共聚焦和Western blot分析结果显示:NaCl胁迫诱导了质膜H+-ATPase的表达,酶蛋白量的增加可能是NaCl胁迫后构树幼苗H+ATPase活性升高的原因之一。

       

      Abstract: This study examined the effects of NaCl stress on the activity and expression of plasma membrane H+-ATPase in roots and leaves of Broussonetia papyrifera. Results showed that plasma membrane H+-ATPase activity of B. papyrifera plantlets increased under salt stress. In roots, the activity was elevated as NaCl concentration increased, while in leaves, it increased at low NaCl concentration and then decreased at high one. Laser confocal microscopy and Western blot analysis demonstrated that the expression of plasma membrane H+-ATPase increased in response to NaCl stress. The elevated protein level of plasma membrane H+ATPase might play a role in the increase of its activity under NaCl stress.
