    Construction of tissue culture breeding system of high yield and quality species in Populus tomentosa

    • ObjectivePopulus tomentosa is mainly used in artificial forest construction, landscape greening and wood processing industries in the northern China. Due to the upgrading and transformation of forestry industry, the original breeding technology of P. tomentosa cannot be suitable to the requirement of high-quality P. tomentosa in recent years. To ensure the extensive application of improved species of P. tomentosa in wood production and urban greening, it is urgent to construct an efficient breeding technology system of P. tomentosa now.
      MethodThree improved forest varieties, ‘Yiyang 1’, ‘Yiyang 2’and ‘Yiyang 3’, were used for rapid breeding system construction using tissue culture technology. This study systematically analyzed the effects of disinfection methods and plant hormones on the acquisition of sterile materials, adventitious buds, rooting and leaf proliferation induction.
      Result(1) The best sterilized method for the three improved poplar varieties was to treat them with 75% ethanol for 30 seconds and 2% sodium hypochlorite for 5 minutes. (2) In adventitious bud induction, 0.3 mg/L 6-BA and 0.1 mg/L NAA were applicable to ‘Yiyang 1’ and ‘Yiyang 2’, while the proliferation coefficient of ‘Yiyang 3’ reached the maximum under the conditions of 0.5 mg/L 6-BA and 0.1 mg/L NAA. (3) In rooting induction, the optimal concentration of IBA of ‘Yiyang 1’and ‘Yiyang 3’ was 0.5 mg/L, and that of ‘Yiyang 2’ was 0.3 mg/L. (4) The results of leaf proliferation showed that the optimal concentrations of plant hormones were 1.0 mg/L 6-BA and 0.1 mg/LNAA in ‘Yiyang 1’ and ‘Yiyang 3’, and 0.7 mg/L 6-BA and 0.1 mg/L NAA in ‘Yiyang 2’.
      ConclusionThe rapid breeding system of ‘Yiyang 1’, ‘Yiyang 2’ and ‘Yiyang 3’ has been preliminarily constructed in this study, which is helpful to promote rapid propagation and genetic transformation research of these improved varieties of P. tomentosa, and lays technical foundation for the directional improvement of improved varieties of P. tomentosa at molecular level.
