    Effects of different concentrations of IBA, NAA on rooting of etiolated shoots of Quercus variabilis and Quercus mongolica

    • ObjectiveThe effects of different concentrations of NAA and IBA on the rooting of one-year-old semi-lignified etiolated shoots of the selected provenances of Quercus variabilis and Quercus mongolica were investigated, aiming to find out what kind of mass concentration is most favorable for the rooting of semi-lignification of the etiolated shoot cuttings of current-year seedlings from fine provenances of Quercus variabilis and Quercus mongolica, and to select the most suitable mass concentration, and provide technical support for the asexual selection of Quercus variabilis and Quercus mongolica.
      MethodIn this study, semi-lignified etiolated shoots of current-year seedlings from selected provenances of Quercus variabilis and Quercus mongolica were used as materials. Two kinds of growth regulators(Butyric acid (IBA) and naphthylacetic acid (NAA))were used. Setting 6 mass concentrations of 3 000, 2 000, 1 500, 750, 600, 400 mg/L, respectively, and rinsing, non-etiolated semi-lignified shoots of the seedlings were used as control to investigate the effects of different concentrations of IBA and NAA on rooting of etiolated shoots of Quercus variabilis and Quercus mongolica.
      ResultThe results showed that the etiolated cuttings were superior to the non-etiolated cuttings in rooting rate, rooting number, root length and root diameter. There were significant differences in adventitious rooting induction between two plant growth regulators for shoot cuttings of Quercus variabilis and Quercus mongolica. When the concentration was 2 000, 1 500, 750, 600, 400 mg/L for IBA, the rooting effects of treated etiolated or non-etiolated shoots of Quercus variabilis and Quercus mongolica were better than those of NAA. Under each mass concentration of growth regulators, the rooting indexes such as the adventitious root incidence and the number of roots of etiolated Quercus variabilis and Quercus mongolica were significantly higher than those of the non-etiolated cuttings. Under the condition of 1 500 mg/L, rooting rate of the etiolated Quercus variabilis was 20.0% higher than that of the non-etiolated one, and the rooting rate of etiolated Quercus mongolica was 14.6% higher than that of the non-etiolated one.
      Conclusion1 500 mg/L IBA is the best plant growth regulator treatment for the etiolated and non-etiolated semi-lignified shoots of the current-year seedlings of Quercus variabilis and Quercus mongolica from superior provenance. The rooting rate of adventitious roots and rooting number were the highest, among which the rooting rate of etiolated Quercus variabilis cuttings was 100%, and that of etiolated Quercus mongolica cuttings was 84.6%.
