Abstract
The stem segments of Acer truncatum were used as explants to study sterilization techniques,browning prevention,proliferation and rooting in vitro.The results show that the optimum season for explant collection is April,the contamination rates of the explants from the 4- and 20-years-old trees are 16.7% and 14.3% after the explants are sterilized in 0.1% aqueous mercuric chloride for 6 min;for the explants from the 4- and 20-years-old trees the addition of PVP of 500 mg/L and 750 mg/L in the medium can prevent browning effectively.The MS medium containing 0.01~0.05 μmol/L TDZ is most efficient in the proliferation of Acer truncatum.The ratio of rhizogenesis is 90.91% in the optimum rooting medium of 1/2 MS medium supplemented with 0.1 μmol/L IBA and under 16 h of photoperiod;the survival ratio of transplanting is 95% after training seedling.
