    菊花叶片不定芽再生体系的研究

    Establishment of shoot regeneration system from leaves of Dendranthema×grandiflora

    • 摘要: 以7个不同菊花品种的叶片为外植体,探讨了基因型、苗龄、叶片着生部位、Vc、活性炭、AgNO3以及不同生长调节剂组合等因素对菊花叶片不定芽再生的影响.试验结果表明,基因型是影响不定芽再生的重要因素;15~35d苗龄的无菌苗中、上部幼嫩叶片不定芽再生能力较高;高浓度NAA有利于提高‘紫妍’叶片外植体的再生能力和消除褐化现象;AgNO3未能促进叶片外植体的再生;适宜‘紫妍’再生的培养基是MS+6-BA 1mg/L+NAA 0.5mg/L和MS+6-BA 2mg/L+NAA 1mg/L;适宜‘L12M57’再生的培养基是MS+6-BA 2mg/L+NAA 0.2mg/L和MS+6 BA 2mg/L+NAA 0.1mg/L.

       

      Abstract: The effects of different factors,such as genotype,seedling age, position of leaves on seedling,Vc,charcoal,AgNO3 and different hormone combinations, on shoot regeneration from leaves of seven different Dendranthema×grandiflora cultivars were studied.Results indicate that the genotype is an important factor in shoot regeneration. The infant leaves from seedlings with an age of 15--35d have high shoot regeneration.High concentrations of NAA accelerate shoot regeneration of Dendranthema×grandiflora ‘Ziyan'.No positive effect of AgNO3 on shoot regeneration is observed.The optimum medium for ‘Ziyan' is MS+6-BA 1mg/L+NAA 0.5mg/L and MS+6-BA 2mg/L+NAA 1mg/L.MS+6-BA 2mg/L+NAA 0.2mg/L and MS+6BA 2 mg/L+NAA 0.1 mg/L is suitable for‘L12M57’.
