Abstract:
In this study we used the seeds of Senecio cruentus ‘Jester’ as explants, and MS as the basal medium to select the optimal combination and concentration of growth regulators for plant regeneration. The results showed that different explants, growth regulator combinations and concentrations had significant effects on plant regeneration. The regernation capacity of laminae was higher than that of petioles. MS + 2.0 mg/L 6-BA gave the best result for multiplication of shoots. The optimal medium for callus multiplication was MS + 1.0 mg/L NAA + 1.0 mg/L 6-BA and for plant regenerated was MS + 2.0 mg/L 6-BA + 2.0 mg/L NAA. For the rooting medium, 1/2 MS added with 0.5 mg/L NAA was the best. Under favorable conditions healthy flowering plants were obtained after transplantation and phenotype test demonstrated that the color of flowers of regenerated plants from different seeds did not change.
