    美丽胡枝子再生体系的研究

    • 摘要: 以美丽胡枝子为材料,研究了6-BA、2,4-D、NAA和苗龄等因素对其子叶节再生、节段扩繁和试管苗生根的影响,目的是要建立其高效再生体系.试验结果表明,6-BA、2,4-D对美丽胡枝子子叶节再生有显著作用,再生最佳培养基为MS+2,4-D 1.0 mg/L+6-BA 0.5 mg/L+NAA 0.5 mg/L;不同苗龄的子叶节再生能力差别不大,但随着苗龄延长,子叶节对6-BA的敏感性增强;6-BA对美丽胡枝子节段不定芽的形成影响也很明显,MS+6-BA 1.0 mg/L+NAA 0.1 mg/L为其最佳增殖培养基;6-BA对美丽胡枝子试管苗的生根作用不大,而NAA对生根的影响显著,最佳生根培养基为MS+6-BA 0.15 mg/L+NAA 0.5 mg/L.

       

      Abstract: In order to establish an effective regeneration system of Lespedeza formosa, the effects of 6-BA,2,4-D,NAA and seedling age on adventitious buds regeneration,proliferation and rooting of L.formosa were studied.The results showed that 6-BA and 2,4-D could evidently accelerate adventitious buds regeneration on cotyledonary node of L.formosa,the optimum regeneration medium was MS+2,4-D(1.0 mg/L)+6-BA 0.5 mg/L+NAA 0.5 mg/L.There were not obvious differences in different seedling ages,but the older cotyledonary node was more sensitive to 6-BA.6-BA affected obviously the differentiation of adventitious buds in segment proliferation,MS+6-BA 1.0 mg/L +NAA 0.1 mg/L was a optimum proliferation medium.It was not 6-BA but NAA that play an important role in rooting,and the optimum rooting medium was MS+6-BA 0.15 mg/L+NAA 0.5 mg/L.
