Abstract:
Using the outer and middle layers of bulb scales as the explants, plant regeneration system of two
Lilium asiatic hybrid-‘Prato’ and ‘Brunello’, was established from culturing on Murashige and Skoog's basal medium supplemented with 6-BA and NAA at different concentrations, and the optimum media for shoot proliferation were:MS medium supplemented with 6-BA 2.0 mg/L and NAA 0.2 mg/L for ‘Prato’, MS with 6-BA 0.5 mg/L and NAA 1.0 mg/L for ‘Brunello’.The differentiation ability for different parts of bulblet leaves was studied, and the results showed that the apical part of leaves did not regenerate, however, mass shoot regeneration occurred from the leaf base, occasionally from the middle part.MS supplemented with 6-BA, NAA and 2, 4-D at different concentrations was carried out to select a suitable method for shoot regeneration.As a result, all the media induced shoot regeneration via direct organoensis, but the medium with 2, 4-D manifested no positive effects.Three kinds of suitable media, using base of bulblet leaves for proliferation were:MS medium supplemented with 6-BA 2.0 mg/L and NAA 0.2 mg/L, or with 6-BA 1.0 mg/L and NAA 1.0 mg/L, or with 6-BA 0.5 mg/L and NAA 0.5 mg/L.The leaf bases which have been separated from the succeeding-cultured mother plantlet for(5-) 6 times could still have a high percentage about 80% of shoot regeneration.
