    Isolation of lipids from photosystem Ⅰ complex in Pisum sativum L.and its characterization with RP-HPLC

    • 摘要: 为进一步探讨脂类物质在类囊体膜及其蛋白复合物中的分布及作用,该研究提取了经DOC-PAGE分离的豌豆光系统Ⅰ中的脂类物质,采用RP-HPLC-UV梯度洗脱分析方法,以乙腈与水为流动相组成,在25 min内进行了较好的分离.所得的色谱图基线平稳、分离度高.RP-HPLC-UV分析图上有6个主要色谱峰,通过所对应的紫外吸收光谱图,确认它们均属于脂类化合物.

       

      Abstract: In order to discuss the distribution and function of lipids in thylakoid membranes and its protein complex,lipids were extracted from photosystem I separated by DOC-PAGE from thylakoid membranes in the leaves of Pisum sativum L.An RP-HPLC-UV method with gradient elution was established for the separation of lipids obtained from thylakoid membranes,and baseline separation of the sample components could be realized in 25 min with this method.From the UV spectrum extracted from the DAD 3D graph,the main components separated in HPLC can be characterized preliminarily as lipids.
