Optimization of purification conditions of polyphenols from Pinus sylvestris var. mongolica using response surface methodology.

Abstract
We optimized the purification process of polyphenols from Pinus sylvetris var. mongolica bark to
provide reference for its development. Static absorption-desorption and dynamic adscorption-desorption
experiment were used to select the best one from nine different types of macroporous resin. Box-Benhnken
design was used to optimize the optimum purification conditions. The optimized purification conditions of
polyphenols from P. sylvetris var. mongolica were: sample solution concentration was 2.5 mg/ mL, 28 mL
sample solution, eluting speed 10 mL/ min, the concentration of ethanol 51.6% and the ratio of diameter
to height of resin column was 1∶27.3. Under the optimized purification conditions, purity of polyphenols
was 69.23%. The results indicate that AB-8 macroporous resin has distinct effect on purification of
polyphenols from P. sylvestris var. mongolica.
